"What-a-ya-mean?" would be used if you didn't understand something that someone said just to make sure that's what he really meant. If you thought that what you said was very clear and didn't need explaining, it would be taken as an insult. In that case, the appropriate reply would be "What-a-ya-mean? What-a-ya-mean?"
